What/How could I make a simple wall to seperate one area of my room from another area?
What materials could I use?
I was thinking about using some rather large/long curtains.

A lot of good suggestions and answers. Depends on the amount of separation and privacy you want to achieve. You can delineate smaller spaces within a larger space with furniture arrangement, that's the most subtle way. A change in color, ceiling or floor height will work, too, but that's an expensive solution in this case. Like you said a curtain, but the screen is a more practical solution. Obviously, building an actual wall will be most private.
I'd go with the screen. You can fold it up, if you want to open the room again.
